The PowerSoil®-htp 96 Soil DNA Isolation Kit is comprised of a novel and proprietary method for isolating genomic DNA from environmental samples in a 96 well format for high throughput. The kit is intended for use with environmental samples containing a high humic acid content including difficult soil types such as compost, marine sediment, and manure among others. The isolated DNA has a high level of purity allowing for more successful PCR amplification of DNA from organisms within the sample. PCR analysis has been performed to detect a variety of organisms including both Gram-positive and Gram-negative bacteria (e.g. Bacillus subtilis, Bacillus anthracis), fungi (e.g. yeasts, molds), algae, and actinomycetes (e.g. Streptomyces) and nematodes.
The PowerSoil®-htp 96 Well Soil DNA Kit distinguishes itself from MO BIO's UltraClean® Soil DNA Isolation Kit with a patented Inhibitor Removal Technology® (IRT), which removes humic substances/brown color. This new procedure is effective at removing PCR inhibitors from even the most difficult soil types.
Environmental samples are added to a 96 well bead beating plate for rapid and thorough homogenization. Cell lysis occurs by mechanical and chemical methods. Total genomic DNA is captured on a 96 well silica membrane in a spin column format. DNA is then washed and eluted from the spin column. DNA is ready for PCR analysis and other downstream applications.
