Description

The DNase Detection Kit* is designed to determine if DNase contamination is present in sample solutions. In the procedure, DNA is incubated with a test samples and is then run on an agarose gel for analysis. The DNase Detection Kit is capable of detecting as low as 10-7 Kunitz units/µl and is ideal for screening PCR buffers and reagents prior to amplification experiments. Shipped on dry ice. Must be kept frozen before use.

Up to four samples can be tested simultaneously with this kit. DNase activity can be quantified by comparison to the reference standards included. The kit is ideal for early detection of DNase activity in reagent solutions that will be used for sensitive DNA based experiments.

The procedure is to mix 1 µl of the test sample with a DNA Ladder standard provided. The sample is incubated at 37°C for 1 hour. After incubation, the sample is loaded onto a native agarose gel along with four control standards included with the kit. Results are interpreted from the gel analysis.

Specifications

The DNase Detection Kit is capable of detecting as low as 10-7 Kunitz units/µl. For clarity, 1 unit of DNase is commonly defined as the amount of enzyme that produces an increase of 1.0 A260 unit at 37°C when DNA is hydrolyzed to acid soluble oligonucleotides.

One Kunitz unit is equal to 50 DNase units. The increase of 1.0 A260 unit represents the loss of
50 µg/ml (0.05 µg/µl) or 1/50 Kunitz unit.
